FOXO4-DRI is a synthetic peptide designed to interfere with the FOXO4-p53 protein-protein interaction that keeps senescent cells alive. FOXO4 (a forkhead box transcription factor) is upregulated in senescent cells and directly binds p53 to sequester it in the nucleus, preventing p53-mediated apoptosis. The DRI peptide competes for this interaction, freeing p53 to trigger apoptosis in senescent cells.
Can I buy FOXO4-DRI for human use?
Research peptide vendors sell FOXO4-DRI for laboratory use only. Self-administration carries risks of contamination (bacterial endotoxins), incorrect dosing, allergic reactions, and unknown long-term effects. It is not approved for human use.
Why haven't FOXO4-DRI human trials started?
Peptide drug development is complex and expensive. FOXO4-DRI requires injectable formulation (no oral bioavailability), immunogenicity testing, and extensive preclinical safety data before human trials. No pharmaceutical company has yet sponsored the required development program.
